Effects of Different Extract Fractions of Xiaoyao San on Behavior and Monoamine Transmitters in Hippocampus of Chronic Unpredictable Mild Stress Rats

Abstract The rat model of chronic unpredictable mild stress was established and the effects of different extract fractions of Xiaoyao San were studied The behavior changes and concentrations of monoamino transmitters (including noradrenaline, 5-hydroxytryptamine and 5-hydroxyindoleacetic acid) in hippocampus of the model rats were investigated after ig administration of the petroleum ether fraction (1), 95% ethanol fraction (2), 1+2 and venlafaxine hydrochloride,
respectively. The behavior changes were evaluated with the results of open-field test and sucrose consumption test as well as the rat weight and the concentrations of three monoamino transmitters were determined by HPLC-ECD. The results showed that the groups of 1 alone and 1 combined with 2 showed the antidepressant effects that were similar to venlafaxine hydroxychloride and three transmitters of the above three group rats were significantly improved. While the effect of 2 alone was not significant, 2 only had a significant up-regulation effect on noradrenaline among the three transmitters.
