    pentamethyl-2-naphthyl)carbonyl]benzoate, which was subjected to Wittig reaction with methyltriphenylphosphonium
    bromide under microwave irradiation and hydrolysis with an overall yield of about 64%(based on compound 2).

    Nitro-containing unsymmetrical diaryl ureas were prepared from 3-nitroaniline and another aryl amine with safe phosgene surrogate triphogene in a one-pot procedure. This protocol features readily available materials, simple operation, short reaction time, good to excellent yield, and no isolation of the reactive isocyanate.

    2-(3-Hydroxy-1-adamantyl)-2-glyoxylic acid, the intermediate of saxagliptin, was synthesized from adamantane via successive reactions with formic acid, thionyl chloride and diethyl ethoxymagnesium malonate to give diethyl 1-adamantaneformyl malonate, which was subjected to hydrolysis and decarboxylation by mixed acid and then oxidation by potassium permanganate. The overall yield was about 28%.

    5-(2-Bromoethyl)-2,3-dihydrobenzofuran, the intermediate of darifenacin, was synthesized from 2,3-dihydrobenzofuranyl-5-acetic acid by reaction with ethyl chloroformate and reduction with sodium borohydride to give 5-hydroxyethyl-2,3-dihydrobenzofuran, which was subjected by bromination with N-bromosuccinimide in the presence of triphenylphosphine with an overall yield of about 83%.

    A medium for Nonomuraea sp. SIPI-D-30 fermentation was developed and the productivity of glycopeptide A40926 B was improved by strain breeding. After SIPI-D-30 was treated with UV and screened in the resistant plate of sodium acetate and glycin, a mutant SIPI-U-157 with higher production of A40926 B was obtained. The effects of carbon and nitrogen sources on the antibiotic production in shake-flask culture were investigated. The optimized medium compositions(g/L) were glucose 5.0, maltodextrin 30.0, soluble starch 10.0, corn starch 30.0, soy peptone 5.0, meat peptone 10.0, yeast extract 5.0, soybean meal 10.0, cotton seed meal 5.0 and L-valine 1.0, in which the production of A40926 B by strain SIPI-U-157 reached 893 mg/L after 7 days' cultivation. In addition, a fed-batch culture of SIPI-U-157 in a 5 L fermentator also conducted. The yield of A40926 B was 583 mg/L at 180 h.

    The fermentation process for the production of ansamitocin P-3(AP-3)by Actinosynnema pretiosum SIPI-3-21 was investigated. By optimization of fermentation medium, the yield of AP-3 reached 123.2 mg/L in the medium containing glucose 20.0 g/L, corn starch 30.0 g/L, cotton seed meal 30.0 g/L, CaCl2 10.0 g/L and CaCO3 5.0 g/L, cultured for 9 days in a shake flask. The effect of isobutanol on AP-3 production was also investigated. The results
    showed that the titer of AP-3 was 307.8 mg/L after feeding 54 mmol/L isobutanol during the fermentation process, and the component of AP-3 in the fermentation broth was increased from 57.2% to 86.5%.

    The anti-inflammatory activity fractions were isolated from Callicarpa kwangtungensis Chun. and purified by column chromatographies, such as AD-8 macroporous resin, Sephadex LH-20, C18 and silica gel. Their structures were identified on the basis of physiochemical properties and spectroscopic methods (1H NMR, 13C NMR, HMBC and ESI-MS). Five C-glycosylflavones were isolated and identified as 5,7,3',4'-tetrahydroxy-8-C-β-Dglucopyranosyl
    flavone(Ⅰ), 5,7,3',4'-tetrahydroxy-6-C-[α-L-rhamnopyranosyl-(1→2)]-β-D-glucopyranosyl flavone (Ⅱ), 5,7,3',4'-tetrahydroxy-8-methoxy-6-C-β-D-glucopyranosyl flavone(Ⅲ), 5,4'-dihydroxy-6,7-dimethoxy-8-C-[β-Dxylopyranosyl-( 1→2)]-β-D-glucopyranosyl flavone(Ⅳ) and 5,4'-dihydroxy-6,7-dimethoxy-8-C-β-D-glucopyranosyl flavone(Ⅴ). CompoundⅠwas isolated from Callicarpa kwangtungensis Chun. for the first time while the others were found from Callicarpa for the first time. The preliminary anti-inflammatory activities of the five compounds were investigated. The results showed that compounds Ⅰ, Ⅱ and Ⅲ had significant in vitro anti-inflammatory activity.

    A tiotropium bromide inhalation solution (TIS) atomized by an Omron vibrating-mesh nebulizer was prepared with atomizing characteristics and stability as the indexes. The aerodynamic particle size distribution (APSD) and fine particle fraction (FPF) of the product nebulized by this device were evaluated by three kinds of equipments, including time of flight (TOF) aerodynamic particle size analyzer, twin-stage liquid impinger and next generation impactor. The stability of the product stored at 60 ℃ or 4 500 lx (4 ℃) for 10 d or 40 ℃ for 3 months was also evaluated. The results showed that the prepared TIS was stable and could provide a consistent delivered dose with a mass median aerodynamic diameter (MMAD) of 2 μm, whose MMAD was similar to the commercial dry powder inhalation. The FPF<5 μm (70.8%) was over twice that of a marketed dry powder inhalation. The results indicated that TIS might be a useful candidate vehicle for the treatment of chronic obstructive pulmonary disease (COPD).

    Astragalus extract powder was pre-granulated with fluidized bed coating to improve the particle size distribution and micromeritics properties, and then  oated with Eudragit E100. The process parameters of pre-granulation were optimized by single factor experiment and orthogonal design. The stabilities of coated granules with different coating level in the accelerated test were investigated with astragaloside Ⅳ content as the index. The results showed that the granules with the coating level over 30% could maintain stable in both high temperature (60 ℃) and high humidity (RH 92.5%) environments.

    SIPI-4884 is a hypolipidemic candidate developed by our lab. The HPLC methods were established for determination of its related substances with the intermediates and possible by-products in the synthesis as the references. For the achiral impurities, a C18 column was used with the mobile phase of tetrahydrofuran-methanol-0.02 mol/L ammonium acetate buffer (adjusted to pH 4.5 with acetic acid) (13︰48︰39) at the detection wavelength of 255 nm. A
    CHIRALCEL AS-H column was used with the mobile phase of hexane-ethanol-TFA (80︰20︰0.1) for the determination of the enantiomer in SIPI-4884. The determination on three batches of SIPI-4884 samples showed that the content of total related substances was less than 1% and the greatest one was the 5-oxo-compound.

    The rheology behavior of methacrylic acid copolymer Ⅰ was investigated and a method was established for the determination of its dynamic viscosity. A Ma lve rn Kinexus Ultra concentric cylinder rheometer was used at 20 ℃ with the shear-rate of 10 s-1. The results showed that methacrylic acid copolymer Ⅰ was a Newtonian fluid, whose dynamic viscosity was 6.78 mPa·s with RSD of 0.19% .

    Danofloxacin residue in bovine muscle was screened by sorbent extraction combined with time resolved luminescence (TRL) directly measured on C18 sorbent strips. The analyte was adsorbed on sorbent surface by immersion in defatted homogenates. After reagent application and desiccation, terbium-sensitized luminescence was directly measured on sorbent surfaces at λex of 273 nm and λem of 546 nm. The calibration curve was linear in the range of
    0 - 1 000 ng/g. Among 48 blind samples randomly fortified at 0 - 1 000 ng/g, 45 samples were screened correctly and 3 negative samples were presumed positive.

    An inductively coupled plasma mass spectrometry method was established for the determination of cadmium, arsenic and lead in vitamin B2 injection. The vitamin B2 injection samples were digested by microwave, with indium, germanium and bismuth as the internal standard elements  and poplar leaves as the controlled reference standard. Both cadmium and lead were linear in the range of 1 - 100 ng/ml, while arsenic was linear in the range of 1 - 150 ng/ml.
    The recoveries were 92.4% - 101.7%, with RSDs less than 4.1%.

    The breakage problem of paroxetine hydrochloride crystallization process had been analyzed and studied in detail with the focused beam reflectance measurement (FBRM®) and Mastersizer particle size analyzer. The results showed that the breakage problem played an important role on crystallization procedure of paroxetine hydrochloride and breakage mechanism was fragmentation breakage. Equal-size breakage model could be used to demonstrate the breakage process. The minimum particle size and metastable zone of breakage process had been proposed for the first time in this paper.

    Tetracyclines are broad spectrum antibiotics which can inhibit protein synthesis and are widely used to treat various bacterial infections. The biosynthesis cluster of tetracyclines belongs to type Ⅱ polyketide synthase (PKS Ⅱ) which generates the poly-β-ketone backbone through successive decarboxylative condensation of malonyl-CoA extender units followed by modifications by cyclases, oxygenases, transferases and additional tailoring enzymes. Genetic
    and biochemical studies have illustrated most of the steps involved in the biosynthesis of tetracyclines, which may be helpful to better understanding and regulation of the biosynthesis of this type of polyketides.

    Five issues related with Grignard reaction: the suitable solvents, the magnesium selection, the magnesium activation, the reaction maintenance, the work-up method and the safety of Grignard reaction have been briefly reviewed. A new and reliable work up method used for hydrolyzing MgX-complex at the end of the reaction was given. Saturated sodium chloride aqueous solution can effectively decompose the MgX-complex to result in a clearly two-phase separation. The new method is superior over the traditional NH4Cl-method, which has been already used for five decades. The method is especially applicable for large scale manufacture.
